Title: Development of microRNA biomarkers for noninvasive early detection of pancreatic cancer

Abstract

Early detection of pancreatic cancer is very difficult because it shows little symptoms in the early stage. It is important to develop specific and noninvasive biomarkers for pancreatic cancer diagnosis, especially for the early-stage tumors. Blood serum samples of 89 pancreatic cancer patients and 20 healthy controls were used in the case-control study. For specificity of marker, we also include blood serum samples of 45 patients with other cancers in control. We identified 189 differentially expressed miRNAs through the logistic regression in FDR 0.01. Among them, we selected 23 miRNA markers through penalized support vector machine of SCAD penalty and build an early diagnosis model for pancreatic cancer using support vector machine. From 100 repetition of 10-fold nested cross validation, average sensitivity and specificity were calculated as 0.95 and 0.92, respectively, which showed greater performance than previous biomarkers for pancreatic cancer diagnosis.
